AC-265347 (AC265347) is a potent, selective CaSR agonist and positive allosteric modulator (ago-PAM), activates CaSR signaling in cellular proliferation and phosphatidylinositol (PI) hydrolysis assays with potencies of 30 and 10 nM, respectively.
AC-265347 significantly stimulates greater responses than Cinacalcet or Calindol at each of four loss-of-function human polymorphic CaSR variants, and does not inhibit the CYP2D6 cytochrome P450 isozyme。
AC-265347 reduces serum parathyroid hormone (PTH) in rats, also reduces plasma ionizable calcium ([Ca(2+)](o)).
