AS2674723 is a potent, selective and brain-penetrating 5-HT5A receptor antagonist with Ki of 0.75, 0.67 and 1.9 nM against human, rat, mouse 5-HT5A receptors, respectively.
AS2674723 displays 10-fold greater selectivity for 5-HT5A over 5-HT7 receptor, and no significant affinity for 5-HT2A/2B/2C/3/4/6 receptors.
AS2674723 (0.01-0.03 mg/kg, p.o.) ameliorated scopolamine-induced working memory deficit in mice and improved reference memory impairment in aged rats.
