Biological Activity
BCT1028 is a potent, selective small molecule Biliverdin IXβ reductase (BLVRB) inhibitor with IC50 of 195 nM (50 uM BLVRB substrate DCPIP).
BCT1028 exhibits high selectivity against a 44-member pharmacologic in vitro target subset.
BCT1028 promotes megakaryocyte speciation in biphenotypic (erythro/megakaryocyte) cellular models and synergizes with TPO-dependent megakaryocyte formation in hematopoietic stem cells.
BCT1028 expands platelet recovery in stress thrombopoietic models with no adverse effects in mice.
Biliverdin IXβ reductase (BLVRB) is an NADPH-dependent enzyme previously implicated in a redox-regulated mechanism of thrombopoiesis distinct from the thrombopoietin (TPO)/c-MPL axis.
