CLOCK inhibitor CLK8 is a CLOCK-binding small molecule disrupting the interaction between CLOCK and BMAL1, interferes with nuclear translocation of CLOCK both in vivo and in vitro.
CLK8 binds in the hollow created between the α2 helix of the bHLH domain and the Hβ strand of the PAS-A domain of CLOCK.
CLK8 alters CLOCK protein levels in a time-dependent manner in U2OS cells.
CLK8 (25 mg/kg, i.p.) enhances the amplitude of the circadian rhythm with no change in period length in treated mice.
