Biological Activity
DCDBS84 hydrobromide is a small molecule specifically targeting c‐Src SH3 RT‐loop with SPR Kd of 0.83 uM, disrupts the interaction of integrin β3 with Src SH3, selectively blocks αIIbβ3 outside‐in signaling and shows potent antithrombotic effect.
DCDBS84 does not affect the tyrosine kinase activity of Src.
DCDBS84 disrupts the interaction of integrin β3 with Src SH3, affecting the phosphorylation of β3 and Src, without affecting canonical PXXP binding.
DCDBS84 selectively inhibits platelet functions mediated by outside‐in signaling.
DCDBS84 effectively suppresses occlusive thrombus formation without affecting bleeding time in mice.
DCDBS84 has potent antithrombotic effect without compromising primary hemostasis by selectively blocking outside‐in signaling through integrin αIIbβ3.
