EB-42486 (EB42486) is a potent, highly selective G2019S-LRRK2 kinase inhibitor with biochemical IC50 of <0.2 nM.
EB-42486 demonstrated cellular potencies ( in HEK293 cell lines overexpressing either WT-LRRK2 or G2019S-LRRK2) for inhibition of pS935-LRRK2 (IC50=3.1 nM), pS1292-LRRK2 (IC50=1.7 nM), and pThr73-Rab10 (11.4 nM), showed 334-, 71-, and 344-fold selectivity over WT LRRK2 respectively.
EB-42486 exhibited impressive kinome selectivity against a panel of 409 kinases, with only 6 kinases being highly inhibited: HPK1, JNK1, JNK2, JNK3, TTK, and YSK4.
