Biological Activity
G4451 is a specific small molecule inhibitor of GPR31, inhibits the GPR31-Gαi3 interaction by targeting the GPR31 conformational transition, effectively blocked MASH progression.
G4451 shows robust inhibitory effects on lipid accumulation, inflammatory response, and MAPK signaling upon PA/OA treatment in primary hepatocytes.
G4451-mediated inhibitory effects were dependent on GPR31 expression.
G4451 (60 mg/kg, or 10 mg/kg/d, orally) effectively blocked metabolic dysfunction–associated steatohepatitis (MASH) in mice and monkeys, greatly suppressed MASH-related genes.
