GBT1118 is a potent, orally bioavailable allosteric modifier of hemoglobin O2 affinity, binds covalently and reversibly via an imine intermediate to the NH2-terminal valine of the Hb α-chain.
GBT1118 increased Hb O2 affinity and decreased the Po2 during hypoxia in awake-instrumented mice.
GBT1118 significantly attenuated bleomycin-induced lung fibrosis, collagen accumulation, body weight loss, and leukocyte infiltration in a bleomycin-induced mouse model of hypoxemia and fibrosis.
GBT1118 significantly improved tolerance to hypoxia in sickle cell mice.
