HGC652 is an E3 ubiquitin ligase TRIM21-dependent molecular glue degrader (TRIM21 Kd=0.06 uM), induces TRIM21-NUP98 interaction and causes selective degradation of nuclear pore complex proteins.
HGC652 induced polyubiquitination of SmF-EGFP-SH3-NUP98APD and SmF-EGFP-PRM in CEBIT assays.
HGC652 also induced the formation of TRIM21-NUP98 complex as shown by the Strep-tag pull-down assays.
HGC652 demonstrated dose-dependent growth inhibition against 8 cancer cell lines with IC50 of 0.106-0.822 uM.
