I194496 (I 194496) is a novel potent, selective inhibitor of cystathionine γ-lyase (CSE) inhibitor with IC50 of 0.79 uM.
I194496 suppressed CSE/H2S system of TNBC cells, significantly inhibited production of H2S in MDA-MB-231 and MDA-MB-468 cells, significantly decreased the protein and mRNA levels of CSE in MDA-MB-231 and MDA-MB-468 cells, but did not affect the expression of CBS protein.
I194496 inhibits the growth, proliferation, migration and invasion of TNBC cells, blocks the PI3K/Akt and Ras/Raf/ERK signaling pathways.
I194496 inhibits the Anxa2/STAT3 and VEGF/FAK/Paxillin signaling pathways in human TNBC cells.
I194496 inhibits human TNBC xenograft tumors in nude mice.
