Isofagomine (Afegostat) is a broad spectrum inhibitor of glycosidases, inhibits beta-glucosidase, glucoamylase, isomaltase, and alpha-glucosidase, specifically and reversibly binds lysosomal hydrolase acid β-glucosidase (GCase) in the ER with high affinity.

Isofagomine (Afegostat) is a broad spectrum inhibitor of glycosidases, inhibits beta-glucosidase, glucoamylase, isomaltase, and alpha-glucosidase, specifically and reversibly binds lysosomal hydrolase acid β-glucosidase (GCase) in the ER with high affinity.
Isofagomine (Afegostat) stabilizes the active form of GCase in the ER and increases trafficking of GCase to lysosomes.
Isofagomine (Afegostat) improved motor and nonmotor function, abolished microglial inflammatory response in the substantia nigra, reduced α-synuclein immunoreactivity in nigral dopaminergic neurons, and reduced the number of small α-synuclein aggregates, while increasing the number of large α-synuclein aggregates.
