JWJ-01-306 is a potent and selective molecular glue degrader of transcription factor ZBTB11 with DC50 of 620 nM, Dmax, 10 µM of 80%, in MIA PaCa-2 cells.
JWJ-01-306 induces otent ZBTB11 degradation and off-target degradation of three related C2H2 ZF transcription factors: ZFP91, ZBTB21 and WIZ, in MIA PaCa-2 cells, but not GSPT1.
JWJ-01-306 promotes formation of the CRBN:molecular glue:ZBTB11 ternary complex.
JWJ-01-306 induces OXPHOS downregulation and reprogramming of mitochondrial metabolism in PDAC cells.
JWJ-01-306 resensitizes KRAS inhibitor-resistant PDAC cells to sotorasib, does not show significantly impaired proliferation alone, counters KRAS inhibitor resistance in multiple types of mutant KRAS-driven PDAC.
JWJ-01-306 reprograms the pancreatic cancer cell transcriptome and proteome to counter the high-OXPHOS drug-resistant state.
