Biological Activity

K-552 is a specific small molecule inhibitor of USP47 with Kd of 12.3 uM (cUSP47CD), shows more specific interactions with cUSP47CD than hUSP7CD.
K-552 inhibits lung cancer cell proliferation in a dose-dependent manner, increases ubiquitinated c-Myc protein level, inhibits cell viability was decreased in four types of NSCLC cell lines, characterized by KRASG12S (A549) and KRASG12C (H358, Calu-1, and HCC1171).
K-552 effectively destabilizes c-Myc and inhibits KRASG12C-mutated NSCLC cell proliferation.
K-552 enhances the efficacy of Sotorasib in vitro and in vivo.
