KB3409 (1 uM) significantly reduced the ROS levels in senescent fibroblasts.
KB3409 interacts with VCP and RCN2.
KB3409 significantly reduces RCN2 mRNA expression in senescent fibroblasts, significantly increases VCP mRNA expression.
KB3409 directly accelerates VCP ATPase activity through an allosteric mechanism of action.
KB3409 promotes mitochondrial quality control by enhancing VCP-associated autophagic flux.
KB3409 decreases mitochondrial Ca2+ overload via RCN2, significantly suppresses mPTP opening in shCTRL-transduced senescent fibroblasts.
KB3409 enhances mitochondrial respiratory capacity, restores mitochondrial function, reduces ROS levels, and functionally reverses cellular senescence phenotypes.
