KC-130 (KC130, ZAK inhibitor 8) is a potent, selective and covalent leucine-zipper and sterile-α motif kinase (ZAK) inhibitor with IC50 of 11.5 nM.
KC-130 displays an extraordinary selectivity against B-raf V600E (IC50>1000 nM), shows kinase selectivity S (10) and S (1) scores are 0.003 and 0.011, respectively, against a panel of 378 wild-type kinases.
KC-130 (0.01-10 uM) dose-dependently inhibited the phosphorylation of p38 in anisomycin-treated U2OS cells.
KC-130 (10 uM) prevented massive cell death observed upon 24 h incubation with doxorubicin.
