Biological Activity
KI-T28-03 is a selective small-molecule TRIM28 inhibitor that binds the TRIM28 PHD-bromodomain with ITC KD of 5 uM for full-length human TRIM28 protein, impairs leukemia cell proliferation and drives myeloid differentiation.
KI-T28-03 stabilizes wild-type TRIM28 but destabilizes the F772A mutant, confirming that F772 is required for productive compound engagement.
KI-T28-03 disrupts TRIM28-dependent programs without globally displacing it from chromatin.
KI-T28-03 inhibits viability of human MOLM13 leukemia cells with IC50 of 4.85 uM.
KI-T28-03 phenocopies key features of genetic TRIM28 loss and selectively impairs leukemia cell fitness.
