KT-333 (KT333) is a potent, highly specific potent heterobifunctional PROTAC degrader of STAT3 with DC50 of 2.5 -11.8 nM in anaplastic T cell lymphoma (ALCL) lines.
KT-333 exhibits selective degradation of STAT3 over nearly 9000 proteins, including other STAT family members.
KT-333 treatment led to rapid apoptosis in tumor cells expressing NPM-ALK fusion oncoproteins.
KT-333 induces a significant reduction in STAT3 and MCL1 protein levels within two weeks of treatment in a cell derived xenograft model of Ven-res AML.
