MC2494 is a novel potent, pan-Sirtuin (SirT) inhibitor, increases RIP1 acetylation in the death domain and induces bona fide tumour-restricted acetylated RIP1/caspase-8-mediated apoptosis.

MC2494 is inactive against both HATs (PCAF) and HDACs, suggesting high pan-SIRT specificity.

MC2494 displays inhibitory actions against SIRT1, SIRT, and SIRT3/4/5/6, revealing unique features when compared with SIRT1- and 2-selective inhibitors.

MC2494 induces caspase-8–dependent cancer-selective cell death.

MC2494 displays broad anticancer action in vitro, ex vivo, and in vivo.

MC2494 exerts cancer-preventive effects in vivo.
