MMRi67 is a potent E3 ligase inhibitor of the MDM2-MDM4 E3 complex, inhibits RING-RING interaction of MDM4 and MDM2B with Kd of 896 nM.
MMRi67 inhibited ubiquitination of every component of the ternary complex, MDM2, MDM4, and p53, in a dose-dependent manner.
MMRi67 induced p53 protein accumulation in NALM6 cells, inhibited leukemic cell proliferation with IC50 of 4.58 uM, by inducing p53-independent apoptosis with low toxicity for healthy peripheral blood monocytes (PBMCs).
