MitoCKi (Creatine kinase inhibitor CKi-26) is a selective, covalent inhibitor of creatine kinases (CKs), inhibits recombinant creatine kinase B (CKB) phosphotransfer activity with IC50 of 185 nM, depletes creatine phosphagen energetics in cells.
MitoCKi (10-20 uM) rapidly and selectively depletes creatine phosphate, and drives toxicity selectively in CK-dependent acute myeloid leukemia.
MitoCKi selectively engages the active site cysteine (Cys283) of CKs in cells. MitoCKi inhibits cell viability in UCSD-AML1 cells with EC50 of 1.8 uM.
