Mufemilast (Hemay005) is a potent, selective, first-in-class PDE4 inhibitor with in vitro IC50 of 80-120 nM, exhibits comparable inhibitory activities to apremilast.
Hemay005 displayed a significant inhibitory effect on TNF-α, which was an important proinflammatory cytokine target in the disease process of psoriasis.
Hemay005 exhibited a considerable efficacy in psoriasis xenograft SCID mouse model compared with apremilast at the same dose of 20 mg/kg per day in the reduction of psoriasis area and epidermal thickness, protein expression changes and
histological scores.
