NCGC00378430 (Compound 8430) is a novel small molecule compound that reduces the SIX1/EYA2 interaction in vitro with IC50 of 52 uM in the Alphascreen assay.
NCGC00378430 partially reversed transcriptional and metabolic profiles mediated by SIX1 overexpression and reversed SIX1-induced TGF-β signaling and epithelial-mesenchymal transition (EMT), also reverses SIX1-induced TGF-β signaling and EMT in vitro in breast cancer cells.
NCGC00378430 had little toxicity in vivo.
NCGC00378430 disrupts the SIX1-EYA2 interaction and transcriptional activity, partially reverses SIX1-mediated transcriptional and metabolic signatures in breast tumor cells.
NCGC00378430 inhibits TGF-β signaling and EMT in breast cancer cells with high SIX1 expression.
NCGC00378430 inhibits metastasis of breast cancer cells expressing high levels of SIX1 in vivo.
