NCGC00241607 (NCGC607) is a small-molecule noninhibitory chaperone of glucocerebrosidase (GCase), successfully chaperoned the mutant enzyme, restored GCase activity and protein levels, and reduced glycolipid storage in neurons.
NCGC607 (3 uM) NCGC607 significantly enhanced GCase activity and increased GCase levels in Gaucher macrophages.
Treatment of Gaucher iDA neurons with NCGC607 increased GCase activity, enhanced translocation of mutant GCase to the lysosome, and reduced substrate levels.
NCGC607 (3 uM) reversed α-Syn accumulation in GD-PD iDA neurons.
