NS5806 is a small molecule cardiac troponin (cTnC) activator, binds to Ca2+-saturated cChimera with Kd of 7.6 uM, stabilizes active troponin, significantly increases the ATPase activity of cardiac myofibrils in a dose-dependent manner.
NS5806 occupies a binding pocket formed by both the Ca2+-saturated NcTnC and cTnISP.
NS5806 strongly increases the calcium sensitivity of force development of demembranated human muscle strips as indicated by an increase in pCa50 by about 0.2 pCa.
NS5806 is a novel calcium sensitizer that stabilizes active troponin.
