Biological Activity

NVL inhibitor MM17 is a small molecule inhibitor of 60S ribosomal subunit assembly factor NVL with IC50 of 0.38 uM, inhibits HCT116 growth in a dose-dependent manner with IC50 of 0.54 uM.
NVL resistance hotspots, particularly H304R and R403W, is sufficient to confer MM17 resistance.
MM17 inhibits NVL and blocks nucleolar 60S biogenesis, has limited impact on 40S production.
MM17 induces p53 dependent and independent cell cycle arrest in HCT116 cells.
