Biological Activity
Np75-4A22 is a specific small molecule inhibitor of the transmembrane domain of death receptor p75NTR (p75NTR TMD), antagonizes NGF-mediated recruitment of the actin-bundling protein fascin to p75NTR, inhibits the migration and lung invasion of human melanoma cells in mice.
Np75-4A22 (1 uM) blocked all NGF activity in Boyden chamber assay of cell chemotaxis.
Np75-4A22 significantly reduced NGF-induced A875 chemotaxis at submicromolar concentrations.
Np75-4A22 inhibited melanoma cell invasion at submicromolar concentrations without inducing cell death.
Np75-4A22 blocked NGF-mediated recruitment of fascin to p75NTR, NGF-induced fascin association with the cytoskeleton and NGF-stimulated filopodia formation.
Np75-4A22 (orally at 200 mg/kg) reduced the spread and seeding of melanoma lung tumors in a mouse xenograft model.
