OSSL_325096 is a novel potent small mlolecule p97/VCP inhibitor, suppresses cell proliferation and induces ER stress and apoptotic cell death in MM cell lines (IC50=0.1-0.5 uM).
OSSL_325096 induces apoptosis in bortezomib‐resistant multiple myeloma (MM) cells and primary MM cells from two bortezomib‐refractory patients but not in PBMC from a healthy donor.
OSSL_325096 inhibits ATPase activity in vitro, downregulates BCL2 and upregulated cell cycle and heat shock‐related proteins.
OSSL_325096 suppressed multiple myeloma (MM) cell proliferation in vivo without lethal toxicity.
