PIT-1 is a specific PIP3 antagonist that disrupts PIP3/Akt PH domain binding with IC50 of 30 uM, Kd of 43.2 uM; inhibits interactions of a number of PIP3-binding PH domains, including those of Akt and PDK1, without affecting several PIP2-selective PH domains; suppresses the PI3K-PDK1-Akt pathway and triggers metabolic stress and apoptosis.
