PTPN22 inhibitor L-1 is a selective, competitive small molecule inhibitor of PTPN22 with IC50 of 1.4 uM, Ki of 0.5 uM, 7- to 10-fold selectivity for PTPN22 over 16 similar PTPs.
PTPN22 inhibitor L1 phenocopied the antitumor effects seen in genotypic PTPN22 knockout.
PTPN22 inhibition via PTPN22 inhibitor L1 promoted activation of CD8+ T cells and macrophage subpopulations toward MHC-II expressing M1-like phenotypes, both of which were necessary for successful antitumor efficacy.
Targeting PTPN22 via PTPN22 inhibitor L1 is nonredundant with and augments anti–PD-1 therapy.
