PXS-4748 hydrochloride (PXS4748) is a potent, selective and irreversible pan-lysyl oxidase (pan-LOX) inhibitor with IC50 of 2.0 uM and 3.2/0.6/1.4/0.2 uM for bovine (aorta) LOX and rh LOXL1/2/3/4, respectively.
PXS-4748 hydrochloride (PXS4748) is a potent, selective and irreversible pan-lysyl oxidase (pan-LOX) inhibitor with IC50 of 2.0 uM and 3.2/0.6/1.4/0.2 uM for bovine (aorta) LOX and rh LOXL1/2/3/4, respectively. PXS-4787 dose-dependently inhibits lysyl oxidases with IC50 values ranging from 0.2 µM (LOXL4) to 3 µM (LOXL1) and displays comparable inhibitory activity across species. PXS-4787 selectivity targets lysyl oxidases and is neither an inhibitor of, nor a substrate for, related oxidases. PXS-4787 (0-10 uM) reduces collagen formation, deposition and crosslinking in primary human dermal fibroblasts cultured in vitro. PXS-4748 effectively target extracellular LOX without impacting on skin cell phenotype or leading to a compensatory response.
