SGC3027 (SGC-3027) is the cell permeable prodrug of SGC8158, a potent, selective, and SAM-competitive inhibitor of PRMT7 with IC50 of <2.5 nM.
SGC3027 inhibited HSP70 methylation with IC50 of 2.4 uM in C2C12 cells.
SGC3027 selectively inhibited PRMT7 but not PRMT1, 4, 5, 6, 9, and DOT1L.
SGC3027 phenocopies the genetic ablation of Prmt7, PRMT7 inhibition via SGC3027 affects cell survival after heat shock or proteasomal stress, sensitized the WT Prmt7 mouse embryonic fibroblasts (MEFs) to bortezomib.
