SIRT7 inhibitor 97491 is a novel specific inhibitor of SIRT7 with IC50 of 0.325 uM, reduces deacetylase activity of SIRT7 in a dose-dependent manner.
SIRT7 inhibitor 97491 inhibits cell proliferation of uterine sarcoma MES-SA cells with IC50 of < 5 uM.
SIRT7 inhibitor 97491 induces p53 acetylation at lysine 373/382 and increases p53 phosphorylation at S392 in treated MES-SA cells (24 h).
SIRT7 inhibitor 97491 promotes apoptosis through caspase pathway and suppresses MES-SA tumor growth in nude mice (20 mg/kg/day, i.p.).
