Salvianolic acid C (YTHDF1 inhibitor SAC) is a selective, competitive small molecule inhibitor of m6A reader YTHDF1 with IC50 of 1.4 uM in in vitro binding assays, shows weaker activity against YTHDF2 (IC50=29 uM), inhibits YTHDF1 and suppresses oligomerization of YTHDF1.
Salvianolic acid C shows issociation constant (KD) against YTHDF1 with 6.3 and 5.3 μM through individual isothermal titration calorimetry (ITC) assay and microscale thermophoresis (MST), respectively.
Salvianolic acid C dissolves YTHDF1 condensates and counteracts hyperactive YTHDF1 in neurons.
Salvianolic acid C reverses neurodevelopmental deficits in fragile X syndrome (FXS) forebrain organoids.
