Sonidegib (Erismodegib, LDE225, NVP-LDE225) is potent, selective, orally available Smoothened (SMO) antagonist, inhibits Hedgehog (Hh) signaling with IC50 of 1.3 nM (mouse) and 2.5 nM (human) in cell-free assays, respectively.
LDE225 treatment also increased sensitivity of ALDH-positive cells to paclitaxel in ovarian cancer.
LDE225 decreased MM cell viability by inducing specific down-regulation of Gli1 and Ptch1, hallmarks of Hh activity.
LDE225 showed antitumor efficacy in combination with bortezomib both in vitro and in vivo in multiple myeloma.
