TC13172 (TC 13172) is a highly potent necroptosis inhibitor targeting the mixed lineage kinase domain-like protein (MLKL), inhibits necroptosis with IC50 of 2 nM in TSZ-induced necroptosis assays in HT-29 cells.
TC13172 (10 uM) does not inhibit the kinase activities of RIP1 or RIP3 in in vitro assays.
TC13172 covalently binds MLKL at Cys-86, blocks the oligomerization and translocation of MLKL from the cytoplasm to the cell membrane.
TC13172 does not disrupt the phosphorylation of MLKL, but decrease the level of MLKL in the membrane phase.
