TREM2 agonist S9 is a potent, small molecule Triggering receptor expressed on myeloid cells-2 (TREM2) agonist with MST binding KD of 0.95 uM.
TREM2 agonist S9 increases SYK phosphorylation in HEK cells expressing TREM2/DAP12, but no in parental HEK cells lacking TREM2/DAP12 expression.
TREM2 agonist S9 engages TREM2 in a manner that preserves signal propagation rather than inducing biased or abortive activation.
TREM2 agonist S9 (1, 5, and 10 µM) modulates Aβ-induced microglial responses in a TREM2-dependent manner in human iPSC-derived microglia, exhibits reduction in IL-1β secretion.
TREM2 agonist S9 preserves synaptic markers in neuron–microglia co-cultures under amyloid stress, exhibits strong suppression of Aβ-induced inflammatory signaling, TREM2-dependent activity, modulation of ApoE secretion, and preservation of synaptic markers.
