Biological Activity
Topobexin is a selective Topoisomerase IIβ (TOP2B) catalytic inhibitor with IC50 of 0.19 uM in DNA decatenation assays, 25-fold selective over TOP2A (IC50=4.8 uM).
Topobexin binds to non-identical residues in the obex pocket. targets the TOP2B ATPase domain.
Topobexin protects cardiomyocytes in isolated rat neonatal ventricular cardiomyocytes (NVCM) against toxicity (LDH release) induced by daunorubicin (DAU).
Topobexin (1 uM) significantly reduced TOP2B-DNA CC in RADAR assays using HEK293F cells, with no effect on TOP2A-DNA CC.
Topobexin (10 mg/kg, i.v.) prevents anthracycline-induced cardiotoxicity in a rabbit model.
