VU0542270 is the first selective inhibitor of vascular Kir6.1/SUR2B KATP channel with IC50 of 129 nM, >300-fold selective over other members of the Kir channel family.
VU0542270 (3 uM) inhibits both Kir6.1- or Kir6.2-containing channels in complex with either SUR2A or SUR2B, but not channels containing SUR1.
VU0542270 shows substantially blunted vasoconstrictive effects, induces ductus arteriosus (DA) constriction in a dose-dependent manner in on isolated mouse DA vessels.
