WJ460 (Myoferlin inhibitor WJ460, WJ-460) is a small molecule targeting myoferlin (MYOF), exerts anti-metastatic activity in the nanomolar range in breast cancer cells with IC50 of 43.37 nM in MDA-MB-231 and 36.40 nM in BT549 cells.
WJ460 significantly suppresses capillary-like tube formation of human umbilical vein endothelial cells (HUVECs), diminishes the level of phospho-VEGFR-2 and total VEGFR-2 in spontaneous metastasis mouse model.
WJ460 abrogates breast cancer lung extravasation and colonization in vivo.
WJ460 directly targets MYOF, binds to MYOF C2 domain (C2D) with Kd of 1.33 uM, facilitates the dissociation of MYOF in Rab7-positive late endosomes, reverses highly metastatic mesenchymal-like breast cancer cells back to an epithelial state and attenuates the metastatic ability in other tumors.
