XEN 602 (XEN602) is a potent, specific divalent metal transporter 1 (DMT1) inhibitor that inhibits Mn incorporation in HEK293 cells with IC50 of 0.3 nM.
XEN 602 decreases p-STAT3 activation, tumor iron accumulation, and tumor cell proliferation.
XEN 602 significantly reduces tumor number and tumor burden, antagonizes the ability of iron to promote tumor growth in CRC mouse model and patient-derived CRC enteroid orthotopic model.
XEN 602 dose-dependently decreases iron absorption in a hyper-absorption mouse model in vivo.
