Biological Activity

cTYK2i Compound-A (Cmpd-A) is a potent, specific and brain-penetrant TYK2 inhibitor (cTYK2i) with IC50 of 6.4 nM.
Cmpd-A is selective for the TYK2 JH2 domain versus all other tested JAK domains
Cmpd-A inhibits IL-6-induced pSTAT3 and GM-CSF-induced pSTAT5 with IC50 of 6.4 nM and 8.0 nM respectively.
Cmpd-A reduced type I IFN pathway activity in the brain.
Cmpd-A is protective by semitherapeutic delivery in the MOG35-55 EAE mouse model, reduces demyelination and gliosis.
Cmpd-A attenuates transcriptional inflammatory program and biomarker expression in EAE mouse models.
